The peyote plants aren't random or spawn at specific circumstances. They spawn just normal. The only peyote in the game that has specificationa is the golden peyote

It only spawns under a certain weather condition and is also only unlocked by achieving 100% game completion

If you followed it in correct order you'll eventually arrive at the scrapyard in Sandy Shores where you fight a werewolf which you can also unlock for Director Mode
