I would say hitting New Game is the only way to start a run. One don't have to worry about loop holes.
Just think about other runs like the HP series
where they always need to watch 2.5 minutes of intro on each reset
Time difference for restarting between New Game and using a save file sound negatable as the game loads fast on PC.

Also I like how it's harder for anyone to cheat if we start from new Game file.

Of course they could splice and such... I don't know if XXL has good anti splicing stuff.

By editing a save file before hand.
So you still have the intro but also some goodies.
And what could you edit anyway ?
